| Abstract :
Confocal microscopy is a fluorescence imaging technique allowing to observe live cells at a resolution of several hundreds of nanometer. The confocal system consists in using a pinhole which removes most of the light coming from out of focus planes.
Even though this dramatically increases the quality of the acquired images, some artifacts remain : blur from the depth direction and Poisson noise due to low-photon detection.
Numerical methods were developed to reduce the blur in such images, they are called deconvolution methods. They require a description of the blur shape, called the point spread function
(PSF).
The purpose of my work is to highlight the need for restoration methods for confocal imaging and present the associated numerical algorithms. |
