Abstract :
A frequent and well-founded criticism of the maximum em a posteriori (MAP) and minimum mean squared error (MMSE) estimates of a continuous parameter param taking values in a differentiable manifold paramspace is that they are not invariant to arbitrary `reparametrizations' of paramspace. This paper clarifies the issues surrounding this problem, by pointing out the difference between coordinate invariance, which is a em sine qua non for a mathematically well-defined problem, and diffeomorphism invariance, which is a substantial issue, and then provides a solution. We first show that the presence of a metric structure on paramspace can be used to define coordinate-invariant MAP and MMSE estimates, and we argue that this is the natural way to proceed. We then discuss the choice of a metric structure on paramspace. By imposing an invariance criterion natural within a Bayesian framework, we show that this choice is essentially unique. It does not necessarily correspond to a choice of coordinates. In cases of complete prior ignorance, when Jeffreys' prior is used, the invariant MAP estimate reduces to the maximum likelihood estimate. The invariant MAP estimate coincides with the minimum message length (MML) estimate, but no discretization or approximation is used in its derivation. |

Abstract :
In this paper, we propose a method for the
iterative restoration of fluorescence Confocal Laser Scanning
Microscopic (CLSM) images and parametric estimation of the
acquisition system’s Point Spread Function (PSF). The CLSM is
an optical fluorescence microscope that scans a specimen in 3D
and uses a pinhole to reject most of the out-of-focus light. However,
the quality of the images suffers from two basic physical
limitations. The diffraction-limited nature of the optical system,
and the reduced amount of light detected by the photomultiplier
cause blur and photon counting noise respectively. These images
can hence benefit from post-processing restoration methods
based on deconvolution. An efficient method for parametric
blind image deconvolution involves the simultaneous estimation
of the specimen 3D distribution of fluorescent sources and
the microscope PSF. By using a model for the microscope
image acquisition physical process, we reduce the number of
free parameters describing the PSF and introduce constraints.
The parameters of the PSF may vary during the course of
experimentation, and so they have to be estimated directly from
the observed data. A priori model of the specimen is further
applied to stabilize the alternate minimization algorithm and to
converge to the solutions. |

Résumé :
Dans ce rapport de recherche, nous rappelons brièvement comment la nature limitée de diffraction de l'objectif d'un microscope optique, et le bruit
intrinsèque peuvent affecter la résolution d'une image observée. Un algorithme de déconvolution aveugle a été proposé en vue de restaurer les fréquences manquants au delà de la limite de diffraction. Cependant, sous d'autres conditions, l'approximation du systéme imageur l'imagerie sans aberration n'est plus valide et donc les aberrations de la phase du front d'onde émergeant d'un médium ne sont plus ignorées. Dans la deuxième partie de
ce rapport de recherche, nous montrons que la distribution d'intensité originelle et la localisation d'un objet peuvent être retrouvées uniquement en obtenant de la phase du front d'onde
réfracté, à partir d'images d'intensité observées. Nous démontrons cela par obtention de la fonction de ou a partir d'une microsphère imagée. Le bruit et l'infl uence de la taille de la
microsphère peuvent être diminués et parfois complètement supprimes des images observées en utilisant un estimateur maximum a posteriori. Néanmoins, a cause de l'incohérence du système d'acquisition, une récupération de phase a partir d'intensités observées n'est possible que si la restauration de la phase est contrainte. Nous avons utilisé l'optique géométrique
pour modéliser la phase du front d'onde réfracté, et nous avons teste l'algorithme sur des images simulées. |
Abstract :
In this research report, we recall briefly how the diffraction-limited nature of an optical microscope's objective, and the intrinsic noise can affect the observed images' resolution. A blind deconvolution algorithm can restore the lost frequencies beyond the diffraction limit. However, under other imaging conditions, the approximation of aberration-free imaging, is not applicable, and the phase aberrations of the emerging wavefront from a specimen immersion medium cannot be ignored any more. We show that an object's location and its original intensity distribution can be recovered by retrieving the refracted wavefront's phase from the observed intensity images. We demonstrate this by retrieving the point-spread function from an imaged microsphere. The noise and the influence of the microsphere size can be mitigated and sometimes completely removed from the observed images by using a maximum a posteriori estimate. However, due to the incoherent nature of the acquisition system, phase retrieval from the observed intensities will be possible only if the phase is constrained. We have used geometrical optics to model the phase of the refracted wavefront, and tested the algorithm on some simulated images. |

Résumé :
Dans ce rapport, nous proposons un modèle markovien a priori structuré à arbre binaire (le TS-MRF) pour la segmentation d'images satellitaires multispectrales. Ce modèle permet de représenter un champ bidimensionnel par une séquence de champs de Markov binaires, chacun correspondant à un noeud de l'arbre. Pour avoir une bonne classification, on peut adapter le modèle TS-MRF à la structure intrinsèque des données, en définissant un MRF, à plusieurs paramètres, très flexible. Bien que l'on définisse le modèle global sur tout l'arbre, l'optimisation et l'estimation peuvent être poursuivis en considérant un noeud à la fois, à partir de la racine jusqu'aux feuilles, avec une réduction significative de la complexité. En effet, on a montré expérimentalement que l'algorithme global est beaucoup plus rapide qu'un algorithme conventionnel fondé sur le modèle markovien d'Ising, en particulier quand le nombre des bandes spectrales est très grand. Grâce à la procédure d'optimisation séquentielle, ce modèle permet aussi de déterminer le nombre des classes présentes dans l'image satellitaire, dans le cadre d'une classification non supervisée, à travers une condition d'arrêt définie localement pour chaque noeud. Nous avons effectué des expériences sur une image SPOT de la baie de Lannion, pour laquelle nous disposons d'une vérité terrain, et nous avons trouvé que le modèle proposé fournit de meilleurs résultats que certains autres modèles de Markov et que d'autres méthodes variationnelles. |
Abstract :
In this work we detail a tree-structured MRF (TS-MRF) prior model useful for segmentation of multispectral satellite images. This model allows a hierarchical representation of a 2-D field by the use of a sequence of binary MRFs, each corresponding to a node in the tree. In order to get good performances, one can fit the intrinsic structure of the data to the TS-MRF model, thereby defining a multi-parameter, flexible, MRF. Although a global MRF model is defined on the whole tree, optimization as well estimation can be carried out by working on a single node at a time, from the root down to the leaves, with a significant reduction in complexity. Indeed the overall algorithm is proved experimentally to be much faster than a comparable algorithm based on a conventional Ising MRF model, especially when the number of bands becomes very large. Thanks to the sequential optimization procedure, this model also addresses the cluster validation problem of unsupervised segmentation, through the use of a stopping condition local to each node. Experiments on a SPOT image of the Lannion Bay, a ground-truth of which is available, prove the superior performance of the algorithm w.r.t. some other MRF based algorithms for supervised segmentation, as well as w.r.t. some variational methods. |

Il est fréquemment dit que les estimées au sens du maximum a posteriori (MAP) et du minimum de l'erreur quadratique moyenne (MMSE) d'un paramètre continu ne sont pas invariantes relativement aux «reparamètrisations» de l'espace des paramètres . Ce rapport clarifie les questions autour de ce problème, en soulignant la différence entre l'invariance aux changements de coordonnées, qui est une condition sine qua non pour un problème mathématiq- uement bien défini, et l'invariance aux difféomorphismes, qui est une question significative, et fournit une solution. On montre d'abord que la présence d'une structure métrique sur peut être utilisée pour définir les estimées aux sens du MAP et du MMSE qui sont invariantes aux changements de coordonnées, et on explique pourquoi cela est la fa on naturelle et nécessaire pour le faire. Le problème de l'estimation et les quantités géométriques qui y sont associées sont tous définis d'une fa on clairement invariante aux changements de coordonnées. On montre que la même estimée au sens du MAP est obtenue en utilisant soit la `maximisation d'une densité' soit une fonction de perte delta, définie de fa on invariante. Puis, on discute le choix d'une métrique pour . En imposant un critère d'invariance qui est naturel dans le cadre bayesien, on montre que ce choix est unique. Il ne correspond pas nécessairement à un choix de coordonnées. L'estimée au sens du MAP qui en résulte coincide avec l'estimée fondée sur la longueur minimum de message (MML), mais la demonstration n'utilise pas de discrétisation ou d'approximation. |

The estimation problem and related geometrical quantities are all defined in a manifestly coordinate-invariant way. We show that the same MAP estimate results from `density maximization' or from using an invariantly-defined delta function loss. We then discuss the choice of a metric structure on . By imposing an invariance criterion natural within a Bayesian framework, we show that this choice is essentially unique. It does not necessarily correspond to a choice of coordinates. The resulting MAP estimate coincides with the minimum message length (MML) estimate, but no discretization or approximation is used in its derivation. |

Résumé :
La déconvolution des images satellitaires floues et bruitées est un problème inverse mal posé. L'inversion directe entraîne une amplification inacceptable du bruit. Généralement, soit le problème est régularisé lors de l'inversion, soit le bruit est filtré après déconvolution et décomposition dans le domaine de la transformée en ondelettes. Nous avons developpé dans ce rapport la deuxième solution, en seuillant les coefficients d'une nouvelle transformée en paquets d'ondelettes complexes, les fonctions de seuillage étant estimées de manière automatique. L'utilisation de paquets d'ondelettes complexes rend cette méthode invariante par translation, et tient compte des directions, tout en restant d'une complexité O(N). Les résultats obtenus présentent à la fois des textures nettes et un très bon rapport signal/bruit dans les zones homogènes. Par rapport aux algorithmes concurrents, la méthode que nous proposons est plus rapide, invariante par rotation, et tient compte de la directionnalité des détails et des textures de l'image pour mieux les restaurer. Les images déconvoluées de cette manière peuvent être utilisées telles quelles (la restauration peut être intégrée directement dans la chaîne d'acquisition). Mais elles peuvent également constituer le point de départ d'une méthode de régularisation adaptative, permettant d'obtenir des contours plus francs. |
Abstract :
The deconvolution of blurred and noisy satellite images is an ill-posed inverse problem. The direct inversion leads to unacceptable noise amplificatio- n. Usually, either the problem is regularized during the inversion process, or the noise is filtered after deconvolution and decomposition in the wavelet transform domain. Herein, we have developed the second solution, by thresholding the coefficients of a new complex wavelet packet transform; the thresholding functions are automatically estimated. The use of complex wavelet packets enables translation invariance, and takes into account the directions, while remaining of complexity O(N). The obtained results exhibit both correctly restored textures and a high SNR in homogeneous areas. Compared to concurrent algorithms, the proposed method is faster, rotation invariant and takes into account the directions of the details and textures of the image to restore them better. The images deconvolved this way can be used as they are (the restoration step proposed here can be directly inserted in the acquisition chain). But they also can provide a starting point of an adaptive regularization method, enabling one to obtain sharper edges. |
